Introduction:
Topotecan (trade name Hycamtin) is a chemotherapeutic agent that is a topoisomerase inhibitor. It is a synthetic, water-soluble analog of the natural chemical compound camptothecin. It is used in the form of its hydrochloride salt to treat ovarian cancer, lung cancer, and other cancer types.
Functions:
1 Topotecan Hydrochloride (Topotecan HCl) is a semi-synthetic derivative of camptothecin. Camptothecin is a natural product extracted from the bark of the tree Camptotheca acuminata. Topoisomerase-I is a nuclear enzyme that relieves torsional strain in DNA by opening single strand breaks. Once topoisomerase-I creates a single strand break, the DNA can rotate in front of the advancing replication fork. Topotecan intercalates between DNA bases. This intercalation disrupts the DNA deplication machinery when it reaches a site where topotecan is intercalated.
2 Topotecan is an antineoplastic agent used to treat small cell lung cancer, and certain cancers of the ovary.
3 Topotecan is also used to treat relapse of small cell lung cancer that initially responded to other drugs. Increases in survival times have been observed in patients treated with topotecan compared to control populations treated with paclitaxel.
